چکیده

In a preliminary report (1) a biotin deficiency in Streptococcus lactis 8039l was found to result, in a greatly diminished ability to convert ornit, hine and carbamyl phosphate to citrulline. In the present investigation a study of the conditions necessary to restore normal activity to biotindeficient, cells has been made. Normal activity is restored to biotindeficient, cells when the cells are incubated for a short time with glucose, phosphate buffer, biotin, and an amino acid supplement which includes glutamine. The requirements for restoration of normal activity and the fact that, this restoration can be inhibited by analogues of purines and pyrimidines or by analogues of amino acids not required in the medium suggest, that synthesis of the enzyme in the presence of biotin is necessary for production of an active enzyme system for the conversion of ornithine to citrulline.

Properties and biotin content of purified preparations of the ornithinecitrulline enzyme of Streptococcus lactis.

Previous investigations have shown that biotin deficiency in Streptococcus lactis 8039 is associated with the loss in ability to convert ornithine and carbamyl phosphate to citrulline (1). Although protein synthesis in the presence of biotin was found to be necessary to restore completely the ornithine-citrulline enzyme activity of the biotin-deficient cells (2), a preliminary report on the bio...

Metabolic Functions of Biotin Iv. the Role of Carbamyl-l-glutamic Acid in the Synthesis of Citrulline by Normal and Biotin-deficient Rats* by Gladys Feldott

A characteristic consequence of a biotin deficiency in animals is a decreased ability to fix C402 into various tissue components including the amino acid arginine (1). Carbon dioxide is incorporated into arginine as a result of the conversion of ornithine to citrulline in the Krebs-Henseleit urea cycle (2). The methods developed by Cohen and Hayano (3, 4) for the synthesis of citrulline from or...

Regulation of arginine-ornithine exchange and the arginine deiminase pathway in Streptococcus lactis.

Streptococcus lactis metabolizes arginine by the arginine deiminase (ADI) pathway. Resting cells of S. lactis grown in the presence of galactose and arginine maintain a high intracellular ornithine pool in the absence of arginine and other exogenous energy sources. Addition of arginine results in a rapid release of ornithine concomitant with the uptake of arginine. Subsequent arginine metabolis...

Amino acid incorporation and biotin deficiency.

Biotin deficiency in chicks has been reported to result in a marked decrease in the synthesis of albumin and in the activity of amylase (3). Kritsman, Konikova, and Yukhnovskaya (4) observed that the incorporation of methionine-S3s into the proteins of biotin-deficient rats was reduced as compared to the normal animals. Studies in vitro gave similar results; also, the addition of biotin to the ...
